Results and Conclusions
An effective and long term method to expand mesenchymal stem cells ex vivo was developed using Dulbecco's Modified Eagle Medium High Glucose media with added patient serum. No external growth factors were added in this novel process.
Results regarding scaffolding material indicate that human bone fragements serve as the best scaffold in comparison to synthetic materials and coral.
3D 384 well hanging drop plates were found to be an excellent means to grow the mesenchymal stem cells into colonies in a 3D environment
An interesting discovery regarding the hanging drop plate’s spheroids was that eventually the cells appeared to be excreting an extracellular matrix of a sort. The excreted substance was present in many examined spheroids. It is hoped that the excreted matrix is either calcium or cartilage as this would indicate that the stem cells differentiated into either osteocytes or chondrocytes respectively. To determine the identity of the structure, we sent spheroids down to pathology to be assessed. We have not yet received results but they will be incorporated into the experiment when we do obtain them.
